Abstract:
A microscope for generating an image of a sample, the microscope includes a light source for generating a pulsed light; an optical scanner receiving the pulsed light and transmitting the pulsed light to the sample to be imaged causing the sample to emit energy; a dispersive element that receives the emitted energy from the sample, disperses the energy into its spectral elements and transmits the spectrally dispersed energy; and a camera that generates a spectrally resolved image of the sample based on the spectrally dispersed energy from the dispersive element. Also described is a method of generating spectrally resolved images of the sample.

Abstract:
A system and method of implementing a two-stage scanning technique with a high-speed microscope. The microscope is operable to provide spectrally resolved, multi-dimensional images from a single scan of a sample. The microscope may include one of a multi-beam point scanning microscope, a single beam line scanning microscope, and a multi-beam line scanning microscope. The sample is first tagged such that, if the sample has a particular makeup, it emits energy at particular wavelengths upon receiving excitation beams. The microscope is used to perform a first, wide area scan. If the sample is determined to have emitted energy having particular characteristics, the microscope performs a second, focused scan of the area that emitted the energy having the particular characteristics. The two-stage scanning technique is automated and may be used to quickly identify rare cells, microbes, viruses, and other components within one or more samples.

Abstract:
A system and method of high-speed microscopy using a two-photon microscope with spectral resolution. The microscope is operable to provide spectrally resolved, multi-dimensional images from a single scan of a sample. The microscope may include one of a multi-beam point scanning microscope, a single beam line scanning microscope, and a multi-beam line scanning microscope. The microscope includes a descanning arrangement such that emitted fluorescence is static on a receiving detector. The detector is a narrow detector with a width at least half the size of the length, to reduce the amount of pixel data being transmitted and improve scan speeds. The microscope may also incorporate one or more binning techniques whereby pixels are binned together to improve resolution or scan speeds.

Abstract:
A system and method of high-speed microscopy using a two-photon microscope with spectral resolution. The microscope is operable to provide two- to five-dimensional fluorescence images of samples, including two or three spatial dimensions, a spectral dimension (for fluorescence emission), and a temporal dimension (on a scale of less than approximately one second). Two-dimensional (spatial) images with a complete wavelength spectrum are generated from a single scan of a sample. The microscope may include one of a multi-beam point scanning microscope, a single beam line scanning microscope, and a multi-beam line scanning microscope. The line scans may be formed using one or more of curved mirrors and lenses. The multiple beams may be formed using one of a grating, an array of lenses, and a beam splitter.
